GHK-Cu is a naturally occurring copper-binding tripeptide composed of glycine, histidine, and lysine (glycyl-L-histidyl-L-lysine) complexed with a copper(II) ion. Found in human plasma, saliva, and urine with concentrations that decline significantly with age, this peptide was first isolated from human plasma in 1973. GHK-Cu functions as a signal and carrier peptide, exerting pleiotropic effects by modulating the expression of thousands of human genes and influencing multiple cellular pathways involved in tissue remodeling and regeneration. Its primary mechanism involves stimulating collagen, elastin, proteoglycan, and glycosaminoglycan synthesis in fibroblasts while activating lysyl oxidase to improve tissue tensile strength. The peptide also suppresses pro-inflammatory cytokines (TNF-α, IL-6) and demonstrates potent antioxidant activity. Clinically, GHK-Cu is widely utilized in skincare and hair products for its ability to improve skin firmness, reduce fine lines and wrinkles, accelerate wound healing, and support overall skin and hair regeneration.
